Effects of MMP-8 Production by LPS and TPCK Stimulation in Fetal Blood Cells

Abstract

|
The purpose of our research was to elucidate the effect of Lipopolysaccharide (LPS) stimulation on fetal blood cells for the Matrix metalloproteinase-8 (MMP-8) production and the effect of Nuclear factor kappa B (NF-kB) inhibitor, Tosylphenylalanine chloromethyl ketone (TPCK) on this system. Cord blood at birth was obtained during the elective cesarean section term and using LPS (1 ¥ìg/mL), stimulated immediately with or without NF-kB inhibitor, TPCK (10 and 20 mM). The levels of MMP-8 were measured with specific immunoassay in the supernatant after 1 through 6 hours. NF-kB activity was measured using Western blotting. LPS stimulation to cord blood significantly raised the level of MMP-8 with the passage of time, while NF-kB inhibitor by TPCK restrained the MMP-8 elevation by LPS stimulation. MMP-8 production by cord blood with LPS stimulation was repressed by NF-kB inhibitor, TPCK. Our data suggests that NF-kB has a regulatory role on MMP-8 production induced by LPS in fetal blood cells.
